Expertise de recherche

La régulation transcriptionnelle est un processus complexe qui exige le recrutement de l’ARN polymérase vers les sites dirigés de transcription, la modification de la structure locale de la chromatine et l’épissage des nouveaux transcrits. Une interruption de la coordination de ce processus, comme la transcription erronée qui active de façon inappropriée des gènes qui modifient le comportement normal des cellules, peut favoriser l’apparition d’un cancer. Dans le but de comprendre clairement la nature de cette anomalie, il est souvent nécessaire d’avoir un aperçu global du système.

Notre équipe de recherche s’intéresse actuellement à l’utilisation des technologies d’avant-garde, à haut débit, comme les biopuces à ADN et le séquençage d’ADN de nouvelle génération afin d’élucider les connexions sous-jacentes à l’activité transcriptionnelle et au comportement des cellules. Pour comprendre ce phénomène dans le contexte de la biologie du cancer, nous utilisons actuellement ces approches pour caractériser les échantillons de patients atteints de la leucémie myéloïde aiguë (LMA) dans le but d’identifier les mutations qui favorisent l’apparition de la maladie. De plus, nous utilisons la levure à fission (Schizosaccharomyces pombe) comme système modèle pour étudier le recrutement de l’ARN polymérase II et son comportement pendant la transcription. Le fait de mieux comprendre la machinerie moléculaire qui est à la base de ces processus nous permettra ultimement de mettre au point de nouveaux médicaments pour traiter le cancer.

Biographie

Brian Wilhelm obtient son diplôme de doctorat en génétique médicale du UBC au Terry Fox Laboratory de Vancouver, en 2003. Il poursuit une formation postdoctorale au Wellcome Trust Sanger Institute, au Royaume-Uni dans le labo du Dr Jurg Bahler, puis au sein du laboratoire du docteur Guy Sauvageau à l’IRIC.

En 2010, Brian Wilhelm a lancé son laboratoire, l’Unité de recherche en génomique à haut débit de l’Institut, et a débuté comme professeur au Département de médecine de l’Université de Montréal.
