Résumé :

An explosive growth in mass spectrometry innovations is contributing to our ability to perform in depth, quantitative, proteome and proteoform analysis.  But no mass spectrometer can detect what is not there.  For this reason, innovative strategies for robust, high throughput, quantitative proteome recovery and purification are essential to the proteomics workflow.  Our lab focuses on detergent-based workflows, which therefore necessitate strategies to purify proteins ahead of mass spectrometry.  Protein precipitation may sound old fashion, but when done properly, it offers the potential for unbiased recovery with the highest protein purity.  Electrophoretic strategies also present unique opportunities for automated protein sample preparation.  Here, we present several approaches to protein sample preparation, applicable to both top-down and bottom-up MS analysis.
