Katherine Borden obtient son diplôme de doctorat en biologie moléculaire de l’Université Yale où elle travaille sous la direction de Fred Richards. Elle effectue ensuite deux stages postdoctoraux au Royaume-Uni, sous la supervision d’Andrew Lane et de Paul Freemont, où elle développe une expertise en utilisation de la résonnance magnétique nucléaire (RMN).

I decided that I really wanted to investigate cancer and try to understand the underline mechanisms with the hope of developing therapeutic strategies for it and at least improving the outcome for patients, probably when I was a post doctorate fellow, when I was doing very basic physical and instructional biology work, which I love and still do in the lab but I really felt the need to do something that was more applicable to society.

Katherine Borden et son équipe tentent de répondre à une question fondamentale en biologie du cancer, soit comment des cellules normales deviennent-elles leucémiques? Leurs travaux de recherche portent sur une protéine spécifique, la eIF4E, et comment des anomalies dans son comportement peuvent se transformer en leucémie. Une meilleure compréhension moléculaire de son processus de dérégulation pourrait être utile dans le développement de thérapies pour guérir la leucémie. D’ailleurs, leurs recherches ont permis de montrer les bienfaits de l’emploi de la ribavirine, un antiviral à large spectre courant, dans le traitement de patients atteints de leucémie myéloïde aiguë. Des essais cliniques sont présentement en cours sur la ribavirine en combinaison avec d’autres agents visant à augmenter la durée et la fréquence des rémissions (www.ribatrial.com).

Biographie

L’intérêt de Katherine Borden pour l’application de la biophysique et la biologie structurelle dans l’étude de problèmes biologiques date de l’époque où elle poursuivait ses études supérieures sous la direction de Fred Richards, à l’Université Yale. Elle continue ses recherches en biologie du cancer à l’aide de la résonnance magnétique nucléaire au Royaume-Uni lors de deux stages postdoctoraux, avec Andrew Lane et Paul Freemont respectivement.

Katherine Borden et son équipe concentrent leurs efforts sur l’élucidation de la base moléculaire du cancer. Ils s’intéressent à la manière dont le dysfonctionnement de certains aspects du métabolisme de l’ARN contribue au développement du cancer dans la cellule. La leucémie les intéresse particulièrement et leurs travaux de recherche intègrent plusieurs disciplines, notamment la biologie moléculaire, la biologie structurelle et la biophysique. Leurs travaux ont mené à l’élaboration d’une thérapie novatrice. Plus précisément, la Katherine Borden et son équipe ont découvert que l’inhibition du facteur eIF4E à l’aide de la ribavirine – un médicament antiviral bien connu – empêche ce facteur de produire des cellules cancéreuses, et ce, sans affecter outre mesure les cellules normales.

Katherine Borden dirige actuellement un projet de recherche clinique, appuyé par la Leukemia and Lymphoma Society (États-Unis), qui réunit des cliniciens de partout au Canada afin de mesurer l’efficacité des traitements de ribavirine chez les patients atteints de leucémie aiguë myéloïde de type M4/M5.

Elle a commencé à étudier le facteur eIF4E dans le cadre de ses premières fonctions universitaires comme professeure adjointe à l’Université de Dalhousie (Halifax). Ses travaux de recherche l’ont ensuite menée à travailler à titre de chercheure et professeure agrégée à la Mont Sinaï School of Medicine à New York et, aujourd’hui, à l’IRIC.
